Recombinant Human Platelet Derived Growth Factor-BB (Yeast)
Description: Recombinant Human PDGF-BB, produced in yeast, is a double, glycosylated polypeptide chain containing 109 amino acids with a molecular mass of approximately 32 Kda.
Source: A DNA sequence encoding the mature human PDGF-B chain protein sequence was obtained from platelet mRNA, sub cloned into an expression vector and expressed in yeast.
First five aminoacids: Ser-Leu-Gly-Ser-Leu
Physical State of the Product: Sterile filtered white lyophilized (freeze-dried) powder.
Formulation: Lyophilized from a 100 mM Acetic Acid solution, without additives.
Solubility: It is recommended to reconstitute the lyophilized PDGF-BB in sterile, distilled water to 0.1 - 1.0 mg/ml, which can then be further diluted to other aqueous solutions.
Purity: greater than 97% as determined by:
-Analysis by reducing and non-reducing SDS-PAGE Silver Stained
Protein Content: Each vial contains 1 mg lyophilized PDGF-BB.
Protein quantitation was carried out by two independent methods:
-Protein assay Quanti-iT Protein Assay Kit for the Qubit fluoremeter (Invitrogen)
-Bradford assay for protein quantification
Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis and Western Blot.
Storage: Store lyophilized PDGF-BB desiccated at 2°-8º C. Following reconstitution, store aliquots at -20º C.
Stability: Lyophilized Platelet-derived Growth Factor BB is stable for at least a year. Upon reconstitution, the preparation is stable for up to 10 days at 2-8º C and greater than 3 month at -20º C. Avoid repeated freeze-thaw cycles.
Molecular Mass: 32 KDa.
Biological Activity: Recombinant Human PDGF-BB is fully biologically active when compared to the NIBSC International Standard (IS) for platelet-derived growth factor-BB, which has a specific activity of 3000 I.U/4ug. The ED50, calculated in a dose-dependant proliferation of Swiss mouse 3T3 fibroblasts assay (measured by 3H-thymidine uptake) is 1-3 ng/ml.
